Kit composition

Steps

1. Please prepare yourself: absolute ethanol, RNase-free 1.5mL centrifuge tube.

2. Remove the washing solution and do the following:

a) Washing solution A: 21 mL was added to 9 mL of absolute ethanol; 42 mL was added to 18 mL of absolute ethanol.

b) Washing solution B: 9 mL was added to 21 mL of absolute ethanol; 18 mL was added to 42 mL of absolute ethanol.

c) If the prepared washing solution is precipitated, it can be dissolved at 37 ° C, shaken and used.

3. Take a RNase-free 1.5 mL centrifuge tube, add 200 μL of sample, mix 4 μL of RNA Carrier, add 300 μL of lysate and 20 μL of digestive solution, vortex for 10 seconds, mix well, and centrifuge at 56 °C for 10 minutes until complete lysis.

4. Add 1000 μL of absolute ethanol, gently mix by inversion, if there is a semi-transparent suspension, does not affect the extraction of RNA and subsequent experiments.

5. Place the adsorption column into the collection tube, transfer 760 μL of the above solution into the adsorption column, let stand for 2 minutes, centrifuge at 12,000 rpm for 4 minutes at 4 ° C, and discard the waste liquid in the collection tube.

6. Transfer the remaining 760 μL of the solution to the adsorption column and repeat step 5.

7. Place the adsorption column back into the collection tube, add 500 μL of Wash A to the adsorption column, centrifuge at 12,000 rpm for 4 minutes at 4 ° C, and discard the waste liquid in the collection tube.

8. Put the adsorption column back into the collection tube, add 500 μL of washing solution B to the adsorption column, centrifuge at 12,000 rpm for 4 minutes at 4 ° C, and discard the waste liquid in the collection tube.

9. Place the column back into the collection tube and centrifuge at 12,000 rpm for 4 minutes at 4 ° C to remove any remaining wash solution.

10 Remove the adsorption column, place it in a new RNase-free 1.5 mL centrifuge tube, add 30-50 μL of eluate, let stand for 3 minutes, centrifuge at 12,000 rpm for 4 minutes at 4 ° C, and collect the RNA solution.

Store at 11.-70 ° C or directly for the next experiment.

Precautions

1. To prevent RNase contamination, it is best to wear disposable clean gloves, a mask, a treated RNase-free container and RNase-free ultrapure water during the experiment.

2. The washing liquid contains irritating chemicals. Please take protective measures to avoid direct contact with the skin and prevent inhalation of nose and mouth. In case of skin or eyes, please rinse immediately with water or saline. Seek medical attention if necessary.
