About half of melanomas carry mutations in the BRAF gene that accelerate tumor cell growth and metastasis. Although most patients benefit from anti-BRAF targeted therapy, treatment resistance and tumor progression are still inevitable.
Vito Quaranta and colleagues studied the response of several BRAF gene-mutated melanoma cells to BRAF inhibitors. Related research results were recently published in the Biophysical Journal, where they found that treated cancer cells entered a previously undiscovered “idle†state—the number of cells produced by division was the same as the number of dead cells.
They developed a mathematical model of drug reaction kinetics to explain how treatment changes the epigenetic state of melanoma cells, leading to this state of zero growth. They speculate that the icy cancer cell population is preparing to acquire new genetic mutations that are resistant to treatment and eventually come back.
They suggest that targeted therapies that inhibit cancer cells in these idle states may delay or even eliminate tumor recurrence more effectively than those that inhibit cancer cells that have already degraded.
